    16 AUG 2017 · Human monoclonal antibody (mAbs) are emerging in the field of cancer therapy and have become an increasing proportion of new drugs that are recently approved. Although there are some methods to obtain antigen-specific mAbs from human B cells, it is generally impossible to directly immunize human beings with antigens of interest. For example, for infectious agents, those approaches are largely restricted. To solve these obstacles, two main approaches have been developed; either by humanizing antigen-specific antibodies from small experimental animals (which is laborious due to the great genetic differences from humans) or rely on the in vitro selection of antigen-specific binders from human antibody repertoires.     16 AUG 2017 · By introducing human immunoglobulin loci (heavy and light chains) into the mouse germlines with inactivated/disrupted endogenous antibody machinery, the transgenic mouse/rat strains can directly generate high affinity, fully human antibodies, which avoid the subsequent costly and tedious in vitro humanization and affinity maturation process. Upon immunization, the Ig transgenes will undergo V(D)J rearrangement, random nucleotide (N region) addition, class switching, and somatic hypermutation to yield impeccable antibodies. According to the vascular heterogeneity and changed microenvironment, tumor vasculatures show multiple alterations and differences with regular tissue vasculatures, such as abnormal vessel wall structures, leakiness to macromolecules and constant formation and/or remodeling of vessels.     16 AUG 2017 · In vivo phage display is a unique high-throughput method for biopanning by which the peptide phage libraries are directly injected into animals for isolating and identifying the phages bound to specific tissues. Compared with other panning methods, the targets used for isolating phage-displayed peptides are more “intact” with the natural molecular structure and are in the functional states in vivo. Those selected peptides can be used as selective protein transduction domains (PTD) or cell-penetrating peptides (CPPs) for transporting chemotherapeutic agents to the specific tissue, thus improving drug specificity and efficacy in reducing toxicity. Furthermore, some selected peptides can block the function of their ligand proteins by themselves. As one typical species in this family, sharks have been around for over 500 million years. Due to survive in marine environment, large quantities of urea are contained in sharks’ blood to prevent dehydration, while also make their antibodies much more robust than that of other vertebrates. Comparing with the various immunoglobulin isotypes produced by other species, sharks, on the other hand, mainly produce IgM, IgW and IgNAR (immunoglobulin new antigen receptor).
